Optimisation of ecotoxical test methods for routine use (ERNTE): development of a test kit with Arthrobacter globiformis (Part 3)

The test includes the effect of pollutants in solution as well as those bound in solids. The test is aimed at impeding the dehydrogenase activity of the test organisms in the sample, with reference to the activity in the control sample. The test systems developed were tested at two sites with a wide contamination spectrum. 

Summary

This project deals with the development of a test kit with Arthrobacter globiformis. The test kit is based on the standardised contact test for whole sediments and soils (DIN 38412 48). 

Procedure and results: 

The contact test was miniaturized and the application of freeze-dried bacteria was studied. 

Now the test provides results within less than six hours and the simple handling emphasises its use for the screening of contaminated soils. For risk assessment, bio tests have to be suitable to distinguish between anthropogenic impacts and the effect of natural factors. This influence is demonstrated by the study of eight different not contaminated soils with various soil characteristics. The results of this study also provide information for the selection of the control and reference soils and can be used to describe the threshold of toxicity for the test system. 

A further part of the present study deals with the practical application of the test system. Therefore two abandoned sites in Hamburg were selected. The first site is an ancient gas work area contaminated with organic pollutants, but also heavy metals, and the second site is a landfill site high-grade contaminated with heavy metals. The results show, that the test kit with Arthrobacter globiformis is suitable for the screening of contaminated land and that it can provide important information for the decontamination.
